{The current Olympic champion, who earned bronze at the World Championships recently, dominated an outstanding victory in one minute 56.53 seconds in what was only her sixth race in a year impacted by a muscle issue.
Compatriot and global silver winner Georgia Hunter Bell took the runner-up spot again in one minute 58.33 seconds, while St Vincent's Shafiqua Maloney (1:58.57) claimed third.
"I've barely raced this year so it was likely less of a struggle for me to compete and want to give it something," stated Hodgkinson.
"The environment out there is crazy so it was extremely enjoyable."
The female-exclusive event packed Icahn Stadium in its second edition, bringing a festive vibe to conventional athletics, with live music and special crowns substituting standard medals.
The tennis legend with 23 major titles Serena Williams, who is wed to the founder of Athlos Alexis O., was present to award the winners with their tiaras.
Last year's winners took home $60,000 (£44,603) - among the biggest cash prizes in track and field.
The event also drew several top athletes, as Kenya's three-time Olympic gold medallist and global record breaker Faith K. triumphed in the mile race, crossing the finish line in four hours 17.78 minutes.
The US Brittany Brown, the 200m bronze medallist from the recent Paris Games, secured the sprint double.
